动脉粥样硬化是一种慢性免疫炎症性疾病,其早期病理特征为血管内皮下异常脂质积聚,进而触发氧化应激及炎症反应,共同推动斑块形成及管腔狭窄。目前动脉粥样硬化已成为全球首要死亡原因,其发病率仍呈加速上升趋势。越来越多的研究表明,即使临床患者的血脂得到有效控制,仍面临较高的心血管事件风险。这一现象凸显了现有临床治疗策略的不足,尤其在氧化应激的临床干预方面存在显著局限性。本综述围绕抗氧化治疗领域尚未满足的临床需求展开,分析基础研究向临床转化过程中的关键挑战,旨在为后续新药研发及突破现有治疗瓶颈提供参考与启示。

Abstract:

Atherosclerosis is a chronic immune-inflammatory disease, its early pathological feature involves abnormal lipid accumulation beneath the vascular endothelium, which subsequently triggers oxidative stress and inflammatory responses, collectively driving plaque formation and luminal stenosis. Atherosclerosis has now become the leading cause of death worldwide, and its incidence continues to show an accelerating upward trend. An increasing number of studies indicate that even when blood lipids in clinical patients are effectively controlled, they still face a high risk of cardiovascular events. This phenomenon highlights the inadequacies of current clinical treatment strategies, particularly their significant limitations in clinical interventions for oxidative stress. This review focuses on the unmet clinical needs in the field of antioxidant therapy, analyzes key challenges in the clinical translation of basic research, and aims to provide insights and guidance for future drug development and overcoming current therapeutic bottlenecks.
